Methotrexate does not primarily affect Foxp3+ regulatory T cells in poly-articular juvenile idiopathic arthritis

چکیده
Materials and methods We sampled 11 patients with active poly-articular JIA (poly-JIA) prior to and 3–6 months after initiating MTX. Moreover, 11 poly-JIA patients in remission on MTX were sampled prior to and 3–6 months after withdrawal of MTX. Frequency and characteristics of Foxp3+CD4+Treg and effector T cell subsets were analyzed by flowcytometry. Function of Treg was evaluated in suppression assays. Responses to human heat shock protein 60 (HSP60) were studied in proliferation assays.

The transcription factor crem-αlpha regulates inflammatory T cell subsets in juvenile idiopathic arthritis
Introduction The cAMP response element (CRE) modulator (CREM)a binds to promoters of genes with CREs and regulates transcription via a chromatin-dependent mechanism. CREMa is important for the T cell pathophysiology of SLE by suppression of IL-2 and CD3ζ but enhancement of IL-17 transcription. Juvenile idiopathic arthritis (JIA) is an autoimmune disease of unknown origin. Th17 cells have a path...

Introduction Juvenile idiopathic arthritis (JIA) is a severe disabling disease affecting paediatric population. 507 children with JIA are monitored in Chelyabinsk region. Oligo-articular and poly-articular types of JIA are often associated with affection of eyes (uveitis). In patients resistant to traditional basic treatment (methotrexate and cyclosporine) F-alpha inhibitors are recommended. Ad...
متن کاملCD39: a regulatory role in childhood arthritis
Background Human regulatory T cells (T reg) are classically defined as CD4+ CD25hi Foxp3+. There is increasing evidence that T cells may be subject to regulation via the conversion of proinflammatory ATP to anti-inflammatory adenosine. Methotrexate, the standard therapy for children with JIA, is proposed to work via this mechanism. CD39 belongs to a family of ectoenzymes that convert ATP into a...
متن کاملCD4+CD25bright regulatory T cells actively regulate inflammation in the joints of patients with the remitting form of juvenile idiopathic arthritis.
This study investigates the role of CD4(+)CD25(+) regulatory T cells during the clinical course of juvenile idiopathic arthritis (JIA). Persistent oligoarticular JIA (pers-OA JIA) is a subtype of JIA with a relatively benign, self-remitting course while extended oligoarticular JIA (ext-OA JIA) is a subtype with a much less favorable prognosis. Our data show that patients with pers-OA JIA displa...
متن کاملCoexpression of CD25 and CD27 identifies FoxP3+ regulatory T cells in inflamed synovia
A better understanding of the role of CD4+CD25+ regulatory T cells in disease pathogenesis should follow from the discovery of reliable markers capable of discriminating regulatory from activated T cells. We report that the CD4+CD25+ population in synovial fluid of juvenile idiopathic arthritis (JIA) patients comprises both regulatory and effector T cells that can be distinguished by expression...
